On Saturday, will teams get 1 runner on the front of the line or 2?

In most races, 15 or more teams, probably only one in the front line.

Are there set positions for the teams on the line, or is it just a first come, first served basis?

Starting positions are selected by ranking within a heat. Teams can pick any location on the starting line in that order. The only exception will be in Girls D3, which was re-seeded on Monday and I don't have the ranked order for the new heats.
